enzyme can not remove the terminal Pro from poly-L-Pro. The enzyme is strictly an aminopeptidase and has no endopeptidase action on polypeptides and proteins. The amino group of the N-terminal amino acid must be free. Di-, tri- and polypeptide substrates are cleaved and the optimum size accomodated by the active center is four residues. The residues cleaved most rapidly are hydrophobic ones
