HEPES uptake of P-glycoprotein substrates is substantially diminished when the HEPES concentration is raised to 25 mM. Addition of 15 or 25 mM HEPES in the uptake buffer results in significant reduction in the accumulation of P-glycoprotein substrates cyclosporine-A, ritonavir, and lopinavir. Bidirectional A-B and B-A transport studies show that permeability ratio in the presence of 25 mM HEPES is significantly higher than control.
