Metals/Ions

Metals/Ions Comment Organism Structure
NaCl when the NaCl concentration increases from 10 mM to 200 mM, positive supercoiling activity of TopR1 decreases. At 300 mM NaCl, TopR1 is no longer able to positively supercoil DNA, but it only relaxes negatively supercoiled DNA. Higher NaCl concentrations clearly inhibits the DNA relaxation catalyzed by TopR1 (400 mM). The increase of the DNA linking number catalyzed by TopR1 is inhibited with increasing salt concentration Saccharolobus solfataricus

negatively supercoiled DNA Saccharolobus solfataricus TopR1 can relax negative supercoils without ATP, while the extent of positive supercoiling catalyzed by TopR1 is dependent on the stoichiometry, the ATP concentration, the NaCl concentration and the assay temperature relaxed DNA

60 90 highest activity based on the amount of positive supercoils introduced into DNA is 90°C. The level of positive superturns introduced by TopR1 diminishes with decreasing temperature. This diminution is regular and is about four supercoils every 5°C. At 60 °C, TopR1 is essentially able to relax the negatively supercoiled substrate Saccharolobus solfataricus
