Catalytic efficacies (kcat/Km) decrease by 2 to 3 orders of magnitude with N-pentanoyl-L-homoserine lactone displaying the highest decrease Bacillus thuringiensis
F64C/F68C in the absence of DTT, the enzymatic activity of F64C/F68C AiiA is impaired as compared to that of the wild-type enzyme, and this reduction is more severe for the long N-octanoyl-L-homoserine lactone substrate than for the short N-pentanoyl-L-homoserine lactone substrate. The kcat value is decreased by 14 and 41fold and the KM by 2 and 4fold, resulting in 21 and 158fold decreases in kcat/KM for the N-pentanoyl-L-homoserine lactone and N-octanoyl-L-homoserine lactone substrates, respectively. The addition of DTT not only partially restores enzyme activity but also recovers the characteristic substrate specificity of the wild-type enzyme Bacillus thuringiensis
