A shuttle expression vector system for Pyrococcus furiosus and Escherichia coli is described allowing the regulated expression of proteins in Pyrococcus. A shuttle vector of Pyrococcus abyssi is redesigned using the overexpression of the HMG-CoA reductase as a selection marker which confers resistance to the antibiotic simvastatin Pyrococcus sp.
