F171L the mutant enzyme of enzyme variant Ib shows lower specific activity than the wild-type enzyme when either kanamycin or its semisynthetic derivative amikacin is used as substrate. Alteration of substrate specificity at 42°C. The acetylating activity for kanamycin is higher at 42°C than at 30°C, the ability to use amikacin as substrate is reduced at 42°C. Escherichia coli cells expressing the mutant enzyme are resistant the amikacin at 37°C but susceptible at 42°C Klebsiella pneumoniae
Y80C the mutant enzyme shows only marginal levels of activity when either amikacin, kanamycin, tobramycin or netilmicin is used as substrate Klebsiella pneumoniae
