The enzyme, found in certain mycobacteria, catalyses the activation of the methylated long-chain fatty acids, mycocerosic acids, by adenylation. The activated acids are transferred to the two hydroxyl groups of phthiocerols and phenolphthiocerols by PapA5, leading to formation of dimycocerosates (DIM) and dimycocerosyl triglycosyl phenolphthiocerol (PGL), respectively.
